PO-2 binds to vitellogenin in plasma samples from sea bream. The antibody cross-reacts well with Vtg in plasma samples from Atlantic salmon (Salmo salar), brown trout (Salmo trutta), rainbow trout (Oncorhynchus mykiss), turbot (Scophthalmus maximus), flounder (Platichthys flesus), Atlantic cod (Gadus morhua), wrasse (Ctenolabrus rupestris), tilapia (Oreochromis niloticus) and carp ( Cyprinus carpio). The degree of cross-reactivity differ between species and with the different methods employed.

Vitellogenin (Vg) is an egg yolk precursor protein expressed solely in female fish and is dormant in male fish, but in the presence of estrogenic EDCs, male fish can express the Vg gene but in a dose dependent manner. The use of Vg gene expression in male fish can be used as a molecular marker of exposure to estrogenic Endocrine Disrupting Chemicals (EDCs).
