靶标

  • 功能Member of the repulsive guidance molecule (RGM) family that performs several functions in the developing and adult nervous system. Regulates cephalic neural tube closure, inhibits neurite outgrowth and cortical neuron branching, and the formation of mature synapses. Binding to its receptor NEO1/neogenin induces activation of RHOA-ROCK1/Rho-kinase signaling pathway through UNC5B-ARHGEF12/LARG-PTK2/FAK1 cascade, leading to collapse of the neuronal growth cone and neurite outgrowth inhibition. Furthermore, RGMA binding to NEO1/neogenin leads to HRAS inactivation by influencing HRAS1-PTK2/FAK1-AKT1 pathway. It also functions as a bone morphogenetic protein (BMP) coreceptor that may signal through SMAD1, SMAD5, and SMAD8.

  • Immunohistochemistical (Formalin/PFA-fixed paraffin-embedded sections) detection of Repulsive Guidance Molecule A antibody (ab26287) in mouse brain. Fixative used: Formaldehyde; Antigen retrieval step: None;  Permeabilization: Yes; Blocking step: 1% Serum for 1 hour @ 25°C. Primary Antibody used at 1/500 incubated for 24 h @ 4°C in 0.1% PBS+ 0.3 Triton X+ 1% goat serum.  This image shows RGM-A expressed in all six layers of cortical neurons of adult mouse brain.

  • IHC-FoFr image of Repulsive Guidance Molecule A staining in rat cortex (left) and hippocampus (right) using ab26287. The sections used came from animals perfused fixed with Paraformaldehyde 4%, in phosphate buffer 0.2M. Following postfixation in the same fixative overnight, the brains were cryoprotected in sucrose 30% overnight.

  • IHC image of Repulsive Guidance Molecule A staining in human Hippocampus FFPE section, performed on a BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab26287, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX
