Chlorotoxin is a chloride channel ligand from the venom of the Egyptian/North African/Israeli scorpion. It blocks small-conductance chloride channels. Glioma cells have recently been shown to express a glioma-specific chloride ion channel (GCC) that is sensitive to chlorotoxin (CTX). Chlorotoxin does not bind to normal brain parenchyma or endothelial cells but specifically binds to tumescent glial cells.
Chlorotoxin is currently (2007) under phase II clinical trials, under the name TM-601 (a synthetic version of chlorotoxin); It crosses blood-brain and tissue barriers and binds to malignant brain tumor cells without affecting healthy tissue.
