    ab81665 is a fluorogenic substrate for ubiquitin hydrolases. Ubiquitin-AMC is an exquisitely sensitive substrate for UCH-L3 (Km = 0.039 µM) and for the Isopeptidase-T (Km = 0.17 - 1.4 µM). Ub-AMC is useful for studying ubiquitin hydrolases when detection sensitivity or continuous monitoring of activity is essential.

     

    Substrate concentrations for assay range from 0.1-1 μM. Typical enzyme concentrations for UCH-L3 are 10-100 pM and for Isopeptidase-T are 10-100 nM. Release of AMC fluorescence by UCH enzymes can be monitored using Ex380 nm and Em460 nm wavelengths, respectively.

It is useful for monitoring deubiquitinating enzymes’ activity or screening inhibitors of deubiquitinating enzymes. The concentration of Ub-AMC used in deubiquitinating reactions is usually 0.1-1 µM. The released AMC fluorescence can be monitored using a fluorometer or plate reader with excitation and emission wavelengths at 360 nm and 460 nm, respectively.
