  • 功能Receptor for brain-derived neurotrophic factor (BDNF), neurotrophin-3 and neurotrophin-4/5 but not nerve growth factor (NGF). Involved in the development and/or maintenance of the nervous system. This is a tyrosine-protein kinase receptor. Known substrates for the TRK receptors are SHC1, PI-3 kinase, and PLC-gamma-1.
  • 组织特异性Isoform TrkB is widely expressed, mainly in the nervous tissue. In the CNS, expression is observed in the cerebral cortex, hippocampus, thalamus, choroid plexus, granular layer of the cerebellum, brain stem, and spinal cord. In the peripheral nervous system, it is expressed in many cranial ganglia, the ophtalmic nerve, the vestibular system, multiple facial structures, the submaxillary glands, and dorsal root ganglia. Isoform TrkB-T1 is expressed in multiple tissues, mainly in brain, pancreas, kidney and heart. Isoform TrkB-T-Shc is predominantly expressed in brain.

Recombinant human TrkB protein (Fc Chimera) 图像

  • Lane 1 – MW markers; Lane 2 – ab83565; Lane 3 – ab83565 treated with PNGase F to remove potential N-linked glycans. 10 µg protein loaded per lane. Drop in MW after treatment with PNGase F indicates presence of N-linked glycans. Band in lane 3 at 35 kDa is PNGase F.
  • A sample of ab83565 without carrier protein was reduced and alkylated and focused on a 3-10 IPG strip then run on a 4-20% Tris-HCl 2D gel. 40 µg protein loaded per lane; Deep Purple™ stained. Spot train indicates presence of multiple isoforms of TrkB FcB Chimera. Spots within the spot train were cut from the gel and identified as TrkB FcB Chimera by protein mass fingerprinting.
  • Densitometry scan, derived from the 2D gel image, demonstrates the purified human cell expressed protein exists in multiple isoforms, which differ according to their level of post-translational modification. The triangle indicates theoretical pI and MW of the protein.
