    G-protein coupled receptor for glutamate. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of down-stream effectors. Signaling activates a phosphatidylinositol-calcium second messenger system and generates a calcium-activated chloride current. Plays an important role in the regulation of synaptic plasticity and the modulation of the neural network activity.

  • ab53090 staining Metabotropic Glutamate Receptor 5 (red) in rat hippocampla neuron by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with formaldehyde, permeabilized with 0.2% Triton X-100 and blocked with 5% serum for 20 minutes at 22°C. Samples were incubated with primary antibody (1/200) for 24 hours at 22°C. An Alexa Fluor® 594-conjugated chicken anti-rabbit IgG polyclonal (1/200) was used as the secondary antibody. Cyan - pre-synaptic marker Bassoon.

    Animals were perfused fixed with 4% paraformaldehyde with 15% of a solution of saturated picric acid, in phosphate buffer 0.1M. Brains were then cut using a cryostat and the immunostainings were performed using ab53090 at a 1/300 dilution.
