    • 相关性Integrins are heterodimeric cell surface receptors composed of alpha and beta subunits, which mediate cell-cell and cell-extracellular matrix attachments. Aberrant integrin expression has been found in many epithelial tumours. Changes in integrin expression have been shown to be important for the growth and early metastatic capacity of melanoma cells. Integrin alpha V beta 3 is the predominant integrin of osteoclasts (bone resorbing cells) and is upregulated in malignant melanoma. Integrin alpha V beta 3 binds to a range of ECM proteins (e.g. vitronectin, fibrinogen, fibronectin) via an Arg-Gly-Asp (RGD) motif.

    Anti-Integrin alpha V beta 3 antibody [27.1 (VNR-1)] 图像

    • Immunofluorescence analysis of U87MG cells, staining Integrin alpha V beta 3 (red) with ab78289 at 1/500 dilution.

      Cells were either untreated (left) or treated with 10 nM PTX for 12 hours (right). Nuclei were stained with DAPI (blue).
    • Overlay histogram showing A431 cells stained with ab78289 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab78289, 2µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.
