Receptor for granulocyte colony-stimulating factor (CSF3), essential for granulocytic maturation. Plays a crucial role in the proliferation, differientation and survival of cells along the neutrophilic lineage. In addition it may function in some adhesion or recognition events at the cell surface.
One or several isoforms have been found in myelogenous leukemia cell line KG-1, leukemia U-937 cell line, in bone marrow cells, placenta, and peripheral blood granulocytes. Isoform GCSFR-2 is found only in leukemia U-937 cells. Isoform GCSFR-3 is highly expressed in placenta.
Belongs to the type I cytokine receptor family. Type 2 subfamily. Contains 5 fibronectin type-III domains. Contains 1 Ig-like C2-type (immunoglobulin-like) domain.
The WSXWS motif appears to be necessary for proper protein folding and thereby efficient intracellular transport and cell-surface receptor binding. The box 1 motif is required for JAK interaction and/or activation.
Immunocytochemistry/ Immunofluorescence - GCSF Receptor antibody [S-1268] (ab18627)This image is courtesy of an anonymous Abreview
ab18627 staining human Bone Marrow cells by ICC/IF. Cells were PFA fixed and permeabilized in 1% Triton X-100 prior to blocking in 0.5% BSA for 1 hour at 25°C. The primary antibody was diluted 1/500 and incubated with the sample for 2 hours at 25°C. A FITC conjugated goat anti-mouse antibody, diluted 1/100, was used as the secondary. The image on the left shows the 40X view of Bone Marrow cells stained for GCSFR showing the FITC stain predominantly on cell membranes. The righthand image is the same image with DAPI overlay to show the cell nuclei.
