This antibody recognizes most of the basic (Type II) keratins. It recognizes keratin polypeptides of 58 and 65 to 67 kD. In keratinized epidermis, 58 kD keratin is present in the basal and suprabasal layers, while 65 to 67 kD keratin is present in the cells above the basal layers. This antibody can be used to stain different cell layers in an epithelial tissue and various neoplasms of epithelial origin.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
This antibody has been pretitered and quality controlled to work on formalin-fixed paraffin-embedded as well as acetone fixed cryostat tissue sections.
No further titration is required.
We suggest an incubation period of 30-60 minutes at room temperature.
However, depending upon the fixation conditions and the staining system employed, optimal incubation should be determined by the user.
Proteolytic digestion of tissue sections is required prior to immunostaining.
In keratinized epidermis, 58 kD cytokeratin is present in the basal and suprabasal layers, while 65 to 67 kD cytokeratin is present in the cells above the basal layers.
