Staining works with methanol/acetone fixation or formaldehyde fixation (not pictured) but methanol/acetone is slightly more reliable. Panel B shows an embryo stained with 1/100 dilution of the primary and Panel A shows an stained embryo in which the primary antibody was omitted. DNA was stained with DAPI (shown in blue).
The signal intensity is relatively weak but it consistently decreases with partial depletion of C.elegans pSMC6 (RPT-4) by RNAi, indicating that this staining is specific to C. elegans RPT-4.
