Lysis buffer used: 1% NP-40 in PBS, supplemented with 0.6 mM PMSF and 6ul/mL protease inhibitor cocktail (Sigma-Aldrich, Inc. catalog # P8340).
Immunoprecipitates were resolved using a16.5% acrylamide gel in tricine-SDS-PAGE. Protein bands were transferred to a nitrocellulose membrane and blocked with Blocking buffer (LiCor Biosciences, part # 927-40000) diluted 1:1 with PBS.
Lane 1 is a negative control using a HEK 293T cell lysate and anti-FLAG antibody. Lane 2 is from a HEK 293T cell lysate. Lane 3 is from a U2OS cell lysate.
The bands shown are at 50, 40, 30, 23 kDa and PDCD5 is at 14 kDa.
