Immunostaining obtained in the rat hippocampus using this antibody diluted 1:1000. Sections come from a perfused-fixed animal. The immunostaining was performed using the `free floating` technique, using direct fluorescence.
I observed some cytoplasmic staining in granular neurons and some neuronal processes (arrows on the figure on the right). The figure shows the staining obtained using the objective X20 (left) in the hippocampus, at the level of granular cells, and right panel : at higher power magnification. The staining is consistent with previous observation of cytoplasmic localisation of Neurofascin in the cytoplasm of hippocampal cells and their axon initial segment (Burkarth N MCN 2007).
The tissues were fixed (animals perfused fixed) with 4% PFA and later postfixed overnight in the same fixative. They were cryoprotected in 30% sucrose and cut using a cryostat.
