Tissues were fixed in 2% PFA for 2 hours and frozen. Six micron sections were cut and blocked with 5% donkey serum, 1% BSA, 0.1% Triton-X & Tween for 1 hour at room temperature. Primary antibody was added at 1:100 dilution (shown) in 0.5% BSA overnight at 4C. We also tested dilutions of 1:250 and 1:500, which also showed positive staining. Secondary antibody, AlexaFluor 594 Donkey anti-Goat, was applied at 1:1000 dilution for 1 hour at room temperature. Nuclei were visualized with DAPI. The ab62817 stained the red pulp of the spleen very brightly and specifically, but we saw no positive staining of macrophages in the mouse liver.
