iPS derived neurons (35 days) were fixed in 4% formaldehyde (10 min) and then incubated in 1% serum, 0.1% triton, 0.1% BSA in PBS for 30 mins to permeabilise the cells and block non-specific binding. The cells were then incubated with primary antibody at 1/200 dilution overnight at 4C. The secondary antibody Donkey anti-sheep IgG H&L Alexa 488 (green) at 1:250 dilution and incubated for 1h at room temp. Total cells were stained with DAPI (blue). Positive alpha synuclein staining was observed (green).
